Lipocalin 2 expression promotes tumor progression and therapy resistance by inhibiting ferroptosis in colorectal cancer

چکیده

Lipocalin 2 is a siderophore-binding protein that regulates iron homeostasis. expression elevated in multiple tumor types; however, the mechanisms drive progression upon remain unclear. When over-expressed, it leads to resistance 5-fluorouracil colon cancer cell lines vitro and vivo by inhibiting ferroptosis. inhibits ferroptosis decreasing intracellular levels stimulating of glutathione peroxidase4 component cysteine glutamate antiporter, xCT. The increase xCT dependent on increased ETS1 over-expressing cells. Inhibiting function with monoclonal antibody decrease chemo-resistance transformation vitro, xenograft mouse models. exhibit positive correlation human samples suggesting pathway we have identified operative samples. These results indicate potential therapeutic target described our study can serve as basis for patients who do not respond chemotherapy.

Lipocalin 2 promotes breast cancer progression.

Here, we report that lipocalin 2 (Lcn2) promotes breast cancer progression, and we identify the mechanisms underlying this function. We first found that Lcn2 levels were consistently associated with invasive breast cancer in human tissue and urine samples. To investigate the function of Lcn2 in breast cancer progression, Lcn2 was overexpressed in human breast cancer cells and was found to up-re...

Histone Methyltransferase SETDB1 Promotes the Progression of Colorectal Cancer by Inhibiting the Expression of TP53

SETDB1 is a novel histone methyltransferase associated with the functional tri-methylation of histone H3K9. Although aberrant high expression of SETDB1 was experimentally obversed in a variety of solid tumors, its underlying mechanisms in human carcinogenesis are not well known. In this study, we investigated the expression of SETDB1 in a large cohort of colorectal cancer (CRC) samples and cell...

Study of FGF14 gene expression and cancer progression in colorectal cancer tissue samples

Background: Colorectal cancer is one of the main causes of cancer death and the third most common malignant cancer worldwide. FGF14 is a member of the large family of fibroblast growth factors. These factors control a wide range of biological functions, including cell proliferation, survival, migration and differentiation that disturbing their expression can lead to cancer. The purpose of this ...

Heme oxygenase-1 promotes tumor progression and metastasis of colorectal carcinoma cells by inhibiting antitumor immunity

Heme oxygenase-1 (HO-1) is upregulated in colorectal carcinoma (CRC) cells. However, the role of HO-1 in the metastatic potential of CRC remains to be elucidated. In this study, we investigated the potential of HO-1 to control the antitumor immunity of CRC. Intercellular adhesion molecule-1 (ICAM-1) plays an important role in the immune surveillance system. Hemin-induced HO-1 expression suppres...

Overexpression of paxillin induced by miR-137 suppression promotes tumor progression and metastasis in colorectal cancer

The deregulation of paxillin (PXN) has been involved in the progression and metastasis of different malignancies including colorectal cancer (CRC). miR-137 is frequently suppressed in CRC. PXN is predicted to be a direct target of miR-137 in CRC cells. On this basis, we hypothesized that overexpression of PXN induced by suppression of miR-137 may promote tumor progression and metastasis and pre...
